I have 1 tile missing & not leaking too much; can this be replaced from inside the Loft? There is no inner membrane, and I have loft access. Any help appreciated, thanks

Unfortunately no it can't be replaced from inside the roof structure,if there slate or tile they have to sit on top of each other and can only be done from the outside,it has to sit on the batterns on the outside and under the tiles,its not possible to do it from the inside.

It is possible, however, to remove a cement tile from inside your loft if there is no roofing felt. You would need to slide the two tiles (which currently lay on the missing tile space) upwards, fit the new one through the gap and slide the two tiles over the replacement tile.
